I will briefly introduce my path from studying processes in passive inorganic to active biological materials. The bulk of the talk will concern 2D and 3D Quantitative Phase Imaging (QPI) to analyze the height, area, and volume fluctuations of cells in epithelial monolayers. I will discuss the two imaging modalities, what information is available from the commercial microscopes (Holomonitor and Tomocube) and how we achieve quantitative data of cell area, height, volume and dry mass fraction. I will discuss the new insights into cell volume regulation and epithelial monolayer dynamics that we get from the novel QPI data sets.
